Adenosylhomocysteine is a competitive inhibitor of S-adenosyl-L-methionine-dependent methyl transferase reactions; therefore adenosylhomocysteinase may play a key role in the control of methylations via regulation of the intracellular concentration of adenosylhomocysteine. Required for DNA methylation-dependent gene silencing.

Mar 8, 2017 S-Adenosyl-l-homocysteine (SAH) is the biosynthetic precursor to homocysteine. SAH is formed by the demethylation of S-adenosyl-l-methionine.

S- (5′-Adenosyl)-L-homocys teine (AdoHcy/SAH) is a component of intracellular homocysteine stress. AdoHcy is a competitive inhibitor (versus AdoMet) of DNA methyltransferases (S-adenosyl-L-methionine (AdoMet)-dependent methyltransferases) involved in epigenetics. However, how cellular metabolism directs circadian chromatin remodeling is virtually unexplored. We report that the S-adenosylhomocysteine (SAH) hydrolyzing enzyme adenosylhomocysteinase (AHCY) cyclically associates to CLOCK-BMAL1 at chromatin sites and promotes circadian transcriptional activity. The method is based on enzymatic conversion of Hcy to S-adenosyl-L-homocysteine (SAH) by the action of SAH hydrolase (SAHase; EC 3.3.1.1), followed by quantification of SAH in a competitive immunoassay with use of a monoclonal anti-SAH antibody.

Due to the donation of methyl group, SAM becomes S-adenosyl-L-homocysteine (SAH). The molecular structure of SAM and SAH was presented in Fig. 2 B. DNA methylation is a reversible process.

2012-04-04 In addition to its unusual mRNA capping and methyltransferase mechanisms, the addition of S-adenosyl homocysteine (SAH), which is the by-product and competitive inhibitor of S-adenosyl methionine (SAM)-mediated methyltransferase reactions, leads to synthesis of poly (A) tails on the 3' end of VSV mRNAs that are 10- or 20-fold longer than normal.
